Research Project:

Owen’s graduate research project is to investigate the relationship between glycooxidation and the thrombolytic cascade in diabetes. Using advanced techniques such as mass spectrometry based multiple reaction monitoring technology, glycooxidative adducts are being identified from our cohort of 151 samples, which are collected from normal and pre-diabetic and diabetic patients. Using global mass spectrometric proteomics approach we found that, methylglyoxal (a byproduct of glycolysis which is elevated in diabetes) modified plasminogen has been found at an arginine residue important to the cleavage of plasminogen. Without this cleavage, the entire thrombolytic cascade can be halted. This methylglyoxal adducted arginine on plasminogen may be adding to the deleterious cardiovascular complications at are seen in diabetes.
